Zeiss Elyra PS.1 Combined TIRF, PALM/STORM and Super-Resolution Structured Illumination Microscope

Elyra

This instrument is a Zeiss Axio Observer.Z1 motorized inverted microscope with the following functionality:

  • Four lasers - 405, 488, 561, and 642 nm, all with sufficient power for STORM (all 100 to 150 mW).
  • Single molecule level Total Internal Refection Microscopy (TIRF) using a 100x/1.46NA oil TIRF objective and Andor 897 Electron Multiplying CCD camera (EM-CCD). 512x512 image format, with 1.6x OptoVar lens in place image pixels correspond to 100 x 100 nm in sample space.
  • Both 2D and 3D PALM/STORM – up to 20 nm lateral resolution, and ~50 nm axial resolution (over a 1.4 micron thickness).
  • Super-resolution structured illumination (SR-SIM) – works with any fluorophore and provides an isotropic two-fold increase in resolution – can be on the order of 100 nm lateral and 220 nm axial resolution.
  • SR-SIM uses a 63x/1.4 NA oil objective and a pco.edge 4.2 sCMOS camera. 2048x2048 sensor with 6.5 micron pixels - with the 1.6x Optovar in place, image pixels correspond to ~65 x 65 nm pixels in the sample plane.
